Fit Equation
The drop-down menus allow you to specify a Parameter, Statistic and Axes.
The Fit Equation drop-down selection determines if Weight is enabled. If you select 4 Parameter Logistic or 5 Parameter Logistic, then you can select 1/Y or 1/Y2.
The 4 Parameter Logistic fit equation is defined as follows:
// Definition: Y=Bottom + (Top-Bottom)/(1+10^((LogEC50-X)*HillSlope))
The 5 Parameter Logistic fit equation will vary depending on the version of iQue Forecyt® software being used.
Prior to iQue Forecyt® 7.0, 5PL fitting equations was:
// Definition: Y=Bottom + (Top-Bottom)/(1+10^((LogEC50-X)*HillSlope))^Symmetry
iQue Forecyt® 7.0 and later, 5PL equation is:
// LogXb = LogEC50 + (1 / HillSlope)*Log((2^(1/S))-1)
// Definition: Y=Bottom + (Top-Bottom)/(1+10^((LogXb-X)*HillSlope))^Symmetry
After you have selected the desired metrics for your Standard Curve, click Ok. The following screen shot shows a Standard Curve for these metrics: Log, (Median, BL3-H of TNF) of TNF.
Note: If your standard curve goes in the opposite direction of what you anticipated, you can go back to the Design tab, select your standard and Click on Edit Standards. You can check Reverse (Series) if it is not checked or un-check it if it is checked.
