Setting Up a Simple Four-Color Experiment for Compensation

 

When you design your experiment designate specific wells in which you will place beads or cells labeled with each dye you’re using in your experiment. If you need to use every well on your plate(s) you can run a separate plate with the compensation beads or cells and apply the compensation to the other plates. It’s a good idea to label the channels in Rename Parameters with the dye you’re using as shown below where we named BL1-H CD4 FITC, BL2-H CD3 FITC, BL3-H PE Dazzle and RL1-H CD8 APC.
