This presentation will describe the generation and application of a dual iQue screening platform to deliver a screen in excess of 500K wells, as well as overview the learning’s from this experience and other screening applications. Dr. Wigglesworth will talk about AstraZeneca’s future plans with the iQue Screener platform and how they envisage being able to use this for extended high throughput screening activities throughout 2016.

 

An Automated HT Flow Cytometry Platform Enabling Measurement of Immune Signaling in Whole Blood for Lead Evaluation

This talk will highlight the need for a whole blood based immune signaling assay and the challenges of developing such an assay. It will describe how the rapid microvolume sampling and automation compatibility of the iQue Screener platform enabled routine screening of ~2000 whole blood samples per day. Finally, the analysis of high content data in ForeCyt Software will be discussed.

 

High-Throughput Screening of Monoclonal Antibodies to Multiplexed Antigens
Dr. Benjamin Hoffstrom, Director of Antibody Technology, Fred Hutchinson Cancer Research Center, USA
The Fred Hutchinson Antibody Technology core lab offers time and cost-efficient custom monoclonal antibody development for in-house and external clients. Since its inception in 1995, the lab has developed over 7500 hybridoma cell lines from mice, rats, and rabbits, several of which are commercially licensed reagents. This talk will highlight the ability of the iQue Screener platform to simultaneously screen for monoclonal antibodies to 5 different antigens. The platform uses a two-stage workflow coupling a combination of target-based hybridoma colony selection and cytometric bead-based target deconvolution. The target identification assay is 25-50 times more sensitive than traditional ELISA-based screens, which allows for rapid ranking of several thousand antibodies based on target affinity and murine isotype. A general overview of the workflow will be outlined and specific projects will be highlighted.
